We produced A and B subunits of PP2A in insect cells and show that CLA induced the formation of an open B’/B heterodimer. The enzymatic activity of the heterodimer was significantly stimulated by CLA treatment. Quantification of the bound heterodimer showed that phosphatase activity was correspondingly increased. Interestingly, CLA did not simply stimulate phosphatase activity, but rather changed the’substrate binding’ mechanism of the PP2A heterotrimer from a catalytic to a non-catalytic mode. This suggests that CLA is able to either stabilize or change the conformation of the B/B heterodimer, producing the ‘open’ heterodimer. Such a conformational change may explain why CLA does not affect PP2A phosphatase activity.
